The strep mutans test procedures:
1. The broth is first activated by adding a bacitracin disk at least 15 minutes before use .
2. Chew paraffin for five minute and collect saliva.   
3. The paraffin is removed and a plastic strip is rotated 10 times over the tongue to become contaminated by saliva.
4. The strip is removed through closed lips to remove excess saliva, and inserted into the tube.
5. Incubation at 37C for 48 hours
6. The strip is removed from the tube and dried at room temperature.
7. The number of adherent colonies are compared with a chart supplied by the manufacturer and given a score.  
A plaque sampling can be taken using a sterile disposable brush tip.  This sample can be applied to a segment of the plastic pallete. Up to 3 such samples may be applied to one plastic strip.  Incubate as in the other method.
