Caco-2 Cell Video

Confluent Caco-2 cells do not express GRP or its receptor (GRP-R). In contrast, both are rapidly up-regulated post-wounding whereupon these proteins direct cell remodeling. In these videos the specific effect of GRP is seen by co-culturing in the presence (left side of screen) or absence (right side of screen) of the antagonist D-Phe6 (bombesin) methyl ester. Prior to GRP/GRP-R up-regulation, antagonist has no effect. But then approximately 4-6 hrs post wounding, these proteins are expressed and antagonist completely blocks cell motility.

ICAM video: Two videos are shown: one of Caco-2 cells post wounding in the presence of rabbit anti-human IgG antibody (Control Antibody Video), and one of cells similarly treated but in the presence of monoclonal mouse anti-human ICAM-1 IgG antibody (ICAM-1 Video).

Confluent Caco-2 cells do not express GRPR or ICAM-1, but both are rapidly up-regulated post-wounding. Note that whereas control antibody has no effect on Caco-2 cell migration into a gap (or wound) created by a sterile razor blade, ICAM-1 antibody progressively attenuates this migration with increasing ICAM-1 expression.
