RESEARCH INTERESTS
Na+/H+ exchangers catalyze the electroneutral exchange of an extracellular Na + for an intracellular H + . These proteins are involved in a variety of physiological processes such as Na + , bicarbonate and water absorption and regulation of pH i , cell proliferation and differentiation. Of nine isoforms identified to date (NHE1-NHE9), NHE1, NHE2, and NHE3 have been described in the human intestinal tract. Our lab is interested in understanding the mechanisms involved in the transcriptional regulation of the human Na + /H + exchangers, NHE2 and NHE3. Using a variety of molecular and biochemical approaches we have cloned the promoter region for both NHE2 and NHE3 isoforms. Since gene regulation can be controlled by transcription factors binding to the specific cis-elements in the promoters, we have identified a number of zinc-finger DNA binding transcription factors that recognize the same DNA motif in NHE2 and NHE3 promoters by mutational analyses and in vitro DNA-protein binding experiments. We are using Chromatin Immunoprecipitation experiments to identify which transcription factors bind these promoters in vivo. In addition, we are interested in understanding the regulatory pathways that modulate the DNA-protein and protein-protein interactions. Furthermore, experiments are in progress to determine the effects of the epigenetic modification of the NHE2 and NHE3 promoters on the transcriptional regulation of these genes and its relationship to the cell and region specific NHE expression.
