Our laboratory is interested in the cellular events that occur during selectin-mediated adhesion. The selectins are expressed on endothelium, platelets and leukocytes, and play a crucial role in leukocyte recruitment during an inflammatory response. Following interactions between endothelial selectins and PSGL-1, a leukocyte selectin ligand, the leukocyte undergoes a massive cytoskeletal rearrangement as it rolls along the endothelial surface. PSGL-1 clusters, associates with the actin cytoskeleton, and migrates to the cellular uropod. These observations have led us to explore the cytoskeletal proteins and signaling pathways that are activated during PSGL-1 mediated leukocyte rolling.
