RESEARCH INTEREST:
How does E.coli transctriptional activator FlhD regulate cell division through induction
of PH regulator CadA? FlhD, transcribed from flhD flagellar master operon, not only is
required to express other flagellar genes, but also reduces cell division in response to
acid stress.By using transposon mutagenesis,which transcriptionally regulated by
FlhD, we found fused gene 97% identity with CadA. Cad operon, induced by low PH,
consists of CadA (encoding lysine decarboxylase), CadB(encoding lysine/cadaverin
antiporter), Cad C(encoding regultor of CadA&CadB).
My approach is:
1. to see whether FlhD directly binds CadA, using gel-shift assay, Dnase footprint,
and in vitro transcription assay.
2. If no direct interaction between FlhD and CadA, use transposon mutagenesis
assay to figure out whether any other factors involve in regulation of cell division by
induction of CadA.
